A
combination of ampicillin and cloxacillin.
It belongs to PENICILLIN group and
exhibits bactericidal action against
a wide range of gram positive and
gram negative bacteria including betalactamase
producing staphylococci.
Most
of the penicillins are inactivated
by an enzyme penicillinase or Beta-Lactamase
producing micro-organism; cloxacillin
is effective against such micro-organisms.
PRESENTATION:
Di-Cillin is available as capsules
(500mg & 250mg) and as Suspension
for children in palatable lemon flavour.
COMPOSITION:
Each 500mg capsules contains:
250mg Ampicillin (asAmpicillin Trihydrate)
250mg Cloxacillin (as Cloxacillin
Sodium)
Each
250mg capsule contains:
125mg Ampicillin (as Ampicillin Trihydrate)
125mg Cloxacillin (as Cloxacillin
Sodium)
Each
5ml suspension contains:
125mg Ampicillin (as Ampicillin Trihydrate)
125mg Cloxacillin (as Cloxacillin
Sodium)
PHARMACOKINETICS:
Ampicillin Trihydrate is relatively
stable in gastric acid and is rapidly
absorbed after oral administration.
It diffuses readily into most body
tissues and fluids. However, penetration
into cerebrospinal fluid and brain
occurs only with meningeal inflammation.
Ampicillin is excreted largely unchanged
in the urine. A high concentration
is reached in bile and a part of it
is excreted in the faeces.
Cloxacillin
Sodium is also stable in gastric acid.
After oral administration absorption
is rapid but incomplete. Once absorbed
it binds to serum protein mainly albumin.
Cloxacillin is excreted unchanged
in urine.
Food can interfere with the absorption
of both Ampicillin & Cloxacillin.
Peak plasma concentration is achieved
about 2 hours following dose of 500mg
by mouth.
INDICATION:
Di-Cillin is abroad spectrum antibiotic
available for the treatment of infections
caused by various gram-positive and
gram-negative bacteria including penicillinase
producing staphylococci. Di-Cillin
is indicated in respiratory tract
infections Ear, Nose and Throat infections.
Skin and soft tissue infections.
Gastro-intestinal infections.
Urinary tract infections.
Pelvic infections and Orthopaedic
infections.
DOSAGE
& ADMINISTRATION:
DI-CILLIN 500MG CAPSULES:
ADULTS:
1-2 capsules 4 to 6 hourly. The dose
can be increased depending upon the
severity of the conditions.

Children:
8-12 years
1-2 capsules 4 to 6 hourly
Di-Cillin
Suspension:
Children:
up to 1 year
½ to 1 teaspoon, 4 to 6 hourly
2-7 years
1 to 2 teaspoon, 4 to 6 hourly
SIDE-EFFECT:
Di-Cillin is generally well tolerated
at recommended dose. Like other penicillins,
side effects are of mild and transitory
nature.
Diarrhoea, nausea, vomiting have occurred
and moderate elevation in serum SGOT
has been noted. Severe pseudomembranous
colitis has also been reported in
some patients. Hypersensitivity reactions
include erythematous and maculopapular
rashes and urticaria. If such rashes
occurs, discontinue the therapy.
CONTRA-INDICATIONS:
Previous hypersensitivity reactions
to any of the penicillins.
PRECAUTIONS:
Prolonged use of antibiotics may give
rise to over growth of non-susceptable
organisms including fungi. If super
infection occurs, appropriate measures
should be taken. In prolonged therapy
and particularly with high dosage
schedules, periodic evaluation of
the renal and haematopoietic systems
is recommended.
